using ethanol as co-solvent, the supercritical CO2 has been put in contact to ethanol in a high- pressure autoclave before entering the extractor. In addition, the condensate (oilresin + ethanol) has been separated using a rotary evaporator (Quimis, Model: 3442). The pressure inside the extractor was measured by a Bourdon-type gauge (0-400 bar, ± 5 bar; Model DIN.S, Wika, Germany) and the temperature was monitored by a thermocouple (NiCr/Ni). 2.5 Chemical Analysis The chemical identification of curcuminoids (expressed in terms of curcumin) in both the extract and residual solid has been performed by spectrophotometry, the curcuminoids (curcumin, desmethoxycurcumin and bis-desmethoxycurcumin) by high-pressure liquid chromatography (HPLC) and the essential oil from the turmeric oilresin has been performed by gas chromatography coupled to mass spectrophotometry (GC-MS). 2.5.1 Chemical Identification of the Curcuminoids by Spectrophotometry The chemical identification of the curcuminoids (computed in terms of curcumin) present in both the oil resin obtained in the extracts and the residual solid material were analyzed by spectrophotometry. 2.5.1.1 Chemical Identification of the Curcuminoids in the Oilresin In order to quantify the curcuminoids in the extracts, the oil resin was diluted in ethanol (pa, Químis) and the samples were analyzed in a spectrophotometer (GBC, Model UV/VVIS 916) at 425 nm. Afterwards, the absorbency (A) was measured and the concentration (C) of curcumnoids in the oilresin expressed in g/ml has been computed using a calibration curve, constructed with standard solutions of curcuminoids (pa., Merck, Darmstadt) diluted in ethanol (pa. Químis) as given bellow.
