2.5.3 Chemical Identification of the Essential Oil in the Oilresin of Turmeric by GC-MS The chemical quantification of the essential oil present in the oilresin from turmeric been performed by performed by gas chromatography coupled to mass spectrophotometer (GC-MS) using the procedure as follow (AOAC, 1984). The extracts obtained by supercritical CO2 extraction have been submitted to hydro-distillation for 2 hours and the essential oil extracted with pentane was injected in a GC-MS system consisting of a gas chromatograph (Varian, Model: 3400) coupled to a mass spectrophotometer (Finnigan, Model: INCOS-XL). The column was a 30 m x 0.245 mm ID fused silica open tubular column coated with 0.25 m (5 %-Phenyl Methylpolysiloxane, J&W Scientific, Model: DB-05). Helium was used as the carrier gas with a linear velocity of 32 cm/s, and the following temperature program was used: Oven temperature 333 K; 333-513 K (4 K/min). The detector temperature was 493 K. The mass spectrophotometer (electron impact at 70 eV) has been operated with a temperature of 453 K. The volatile compounds in the essential oil have been identified by comparing their mass spectrum and retention indices with those of standard substances available in the reference library inside the GC-MS system (Jennings, 1980; Adams, 1995). 3. RESULTS AND DISCUSSIONS The operating conditions and process parameters of the supercritical CO2 extraction of oil- resin from the rhizomes of turmeric (Curcuma Longa L) are shown in Table 1. Table 1. Process parameters and operating conditions for the extraction of oilresin from the rhizomes of turmeric (Curcuma longa L) with supercritical CO2 at 318 K and supercritical CO2 using ethanol as co-solvent at 313 K.
